Alpha amylase is classified as an endo-acting carbohydrase enzyme (EC 3.2.1.1). Unlike enzymes that remove sugars from chain ends, it attacks internal α-1,4 glycosidic bonds — rapidly reducing starch into smaller carbohydrate fragments and lowering viscosity during processing.

Alpha amylase binds to starch molecules, recognizes internal α-1,4 glycosidic bonds, and hydrolyzes them using water — producing dextrins and smaller carbohydrates. Because it acts internally within starch molecules, it rapidly reduces starch viscosity. Further breakdown into simple sugars may occur through additional enzymes.

Most commercial food-grade alpha amylase is produced through microbial fermentation using Bacillus licheniformis, B. amyloliquefaciens, B. subtilis, Aspergillus oryzae, and A. niger. It also occurs naturally in humans (salivary glands, pancreas), plants during seed germination (barley, wheat, rice, corn), and fungi.
